Klotho engineered extracellular vesicle in chronic kidney disease preclinical models
Chronic kidney disease, a progressive form of acute kidney injury characterized by renal dysfunction, has a high morbidity and mortality rate related to accelerated aging and cardiovascular disease1. Extracellular vesicles (EVs) are membranous cargo particles that carry nucleic acids, proteins, organelles, and other biomolecules among cells2. Depending on the surface proteins and cargo content, EVs can modulate cell metabolism and the fate of the target cell. In laboratories and clinics, EVs are used in different fields: biomarker discovery, diagnosis, drug delivery and disease therapy. Prof. Bussolati's group found that urinary EVs carrying Klotho protein recovered kidney function in mouse models with acute tubular injury3. In this study, we aimed to engineer EVs by a Klotho peptide and investigate its potential therapeutic effects in chronic kidney disease preclinical models.
