Salivary extracellular vesicles microRNAs profiling as suitable non-invasive source for biomarkers in oral cancer.
Concise Summary
Several studies investigated the expression of micro RNAs (miRNAs) in saliva as potential biomarkers. Since miRNA associated with extracellular vesicles (EVs) are known to be protected from enzymatic degradation, we firstly evaluated whether salivary EVs of patients with oral squamous cell carcinoma (OSCC) are enriched of selective subsets of miRNAs.
Total RNA was extracted from salivary EVs from OSCC patients and healthy controls. The differential expression of miRNAs was evaluated by microarray and later by qRT-PCR. The discrimination power as biomarkers of miRNAs up-regulated in OSCC patients versus controls was evaluated by the Receiver Operating Characteristic (ROC) curves.
We identified a subset of six miRNAs differentially expressed between OSCC patients and controls. Of these, miR-302b-3p and miR-517b-3p were only found in patients' EVs, while miR-512-3p and miR-412-3p were upregulated in patient's EVs and the ROC curve showed a good discrimination power for OSCC diagnosis; miR-27a-3p and miR-494 were overexpressed in OSCC patients, but the ROC curve showed a lower discrimination power. The Kyoto Encyclopedia of Gene and Genomes pathway analysis showed that the six miRNAs are also suggestively involved in pathway activated in OSCC.
In this present work, for the first time to the best of our knowledge, we confirmed the possibility of using salivary EVs as a non-invasive source of miRNAs for OSCC diagnosis and we identified a subset of miRNAs selectively enriched in EVs of OSCC patients that may be used as possibly biomarkers.
