During the first two years of my PhD, my activity was dedicated to the study of lymphoptoliferative diseases with a focus on High Grade B-cells Lymphomas.

These are extremely heterogeneous neoplasm: our goal is to identify a cytogenetic/molecular algorithm in Aggressive B-cells Lymphomas that considers the new “-omics” methods (NGS sequencing, gene expression profiling), to reach a better risk stratification of patients.

We have concluded the retrospectives enrolment of cases with an Aggressive B-cell Lymphoma with MYC rearrangement and/or MYC copy number anomalies (identified by FISH) and treated with Rituximab-CHOP like regimens with curative intent.

The cohort is composed of forty-two patients diagnosed from 1st January 2012 to 31th December 2019 in hematological centers of “Rete Oncologica del Piemonte e della Valle d‘Aosta” with Large B-Cell Lymphoma with MYC abnormalities were enrolled.

On these cases we have carried out the following molecular and cytogenetic analyses:

• Fluorescent In Situ Hybridization (FISH) to determine MYC, BCL2 and BCL6 rearrangements, MYC partner gene, MYC copy number and TP53 Deletion.
• Gene Expression Profiling Study to determine COO.

On the same cases we will carry out also Next Generation Sequencing mutational analysis and a detailed correlation with clinical data to provide a further insight.

At the same time at this project, it was also possible to collaborate with other projects carried out by my reference laboratory (Experimental Pathology – Candiolo Institute – IRCCS, acquiring skills in the molecular pathology field.
